I have submitted a few 5 seconds length video, in Product Page Optimization/ Header
1 day has passed, it seems like the message "The poster frame for this file is processing." still appearing.
The Product Page Optimization has submitted and pending for review.
Is such a message "The poster frame for this file is processing." a concern?

Same issue here. However, mine are with two (of three) 25+ second videos. One of the three behaved normally. The second uploaded and processed normally, allowed me to select a poster frame, and has been processing that poster frame ever since. The third video never allowed me to select a poster frame.
All three videos are tied to an app version update currently “waiting for review.” Worried it is going to hold things up.

I'm only able to edit it once; after I set the poster frame, it just says processing and stays like that for several days.
